Navigating Uncertainty in Pharmacogenomics: Laboratory Challenges and Variant Interpretation

1. Explain the clinical and familial implications of undetected DPYD variants and 5-FU toxicity risk.
2. Identify challenges and gaps in standard pharmacogenomic testing, including the limitations of current variant detection and interpretation.
3. Describe how to overcome laboratory challenges in implementing pharmacogenomic testing.
4. Evaluate innovative research strategies such as experimental saturation mutagenesis, AI-based functional prediction, and large-scale biobank studies, to improve the interpretation of pharmacogenetic variants, including variants of uncertain significance (VUS).
5. Discuss implementation barriers and solutions for pharmacogenomic testing in diverse clinical and laboratory settings, drawing on real-world examples.
